In the past 15 years, mammalian cell culture production processes have moved from the use of glass and stainless-steel bioreactors to disposable, single use bioreactor systems.

Thermo Fisher Scientific’s HyPerforma SUB systems, with scalable volumes ranging from 50L – 2000L is designed using traditional stainless-steel bioreactor principles to ensure optimal cell culture performance. This 2-day course will cover the fundamentals of Single Use Technology (SUT), sensors technology, assemblies and film material, mammalian cell expansion and scale up, the different modes of bioreactor operation and also installation, calibration and use of the HyPerforma SUB.

At the end of the course, participants will be fully equipped with the relevant skillsets to be part of the operations arm of the biomanufacturing industry.

What You Will Learn

Theory Fundamentals of Single Use Technology (SUT)
  • Introduction to SUT and its applications
  • Understand the major differences between SUT and conventional processes in terms of logistics, cost, turnaround time and operation
Sensors technology, assemblies and film material
  • Understand the function of each components associated with SUT
  • Assembly and operation of single use bioreactor systems
Cell expansion and scale up
  • Understand major culture scale up challenges and criteria in terms of media requirement, O2 supply, pH control and mixing
  • Determine operating condition at large scale culture using empirical data from lab and pilot scale
Batch, fed batch and perfusion culture
  • Understand different culture modes and their applications
  • Determine yield, productivity and cost analysis of bioprocess
Practical Bioreactor bag installation
  • Handling, fitting into holder, inflating and connection assembly
Integrity testing
  • Using integrity tester
Aseptic transfer
  • Test run using water, inoculation and sampling
Calibration of sensors
  • Dissolve oxygen, pH and pump calibration
Process control
  • Set-points, feeding and analysis of historical run data
